Chinese state media say the new virus outbreak is the worst since Wuhan
Nearly 200 people have been infected since the virus was first detected on July 20 at the city's busiest airport.
All flights from Nanjing Airport will be grounded until August 11, local media reported.
The city's 9.3 million people - including visitors - will be tested, according to the state - controlled Xinhua news agency.
Posts on social media showed people standing in long queues, and officials reportedly urged people to wear masks and stand at a distance of one meter and avoid talking while they waited.
Officials said the highly contagious delta variant of the virus was behind the infections, adding that the cases were further spread because of how busy the airport was.
Ding Jie, a health official in Nanjing, told reporters the cases came to the city on July 10 with cleaners working on a flight from Russia.
Xinhua News reported that the cleaners did not follow strict hygiene measures.
Airport management has denounced the Communist Party's senior disciplinary body for "problems such as lack of oversight and professional management."
The test now shows that it is spread over at least 13 cities, including Chengdu and the capital Beijing.
However, experts quoted by the Global Times said they believe the eruption is still in its infancy and can be controlled.
Local officials in Nanjing said the condition of the seven victims was Critical.
The new spike in lawsuits has led to speculation on Chinese social media as to whether Chinese vaccines work against delta varient.
It is not clear whether the victims were vaccinated.
Many Southeast Asian countries that rely on Chinese vaccines have recently announced the use of other jabs.
By closing borders and moving quickly to prevent local explosions, China has so far been able to control most of the virus.
